Abstract

The mixed leukocyte culture (MLC) test has come into increasing use in the last few years, both in experimental studies and as a clinical test in matching potential donors and recipients for transplantation. It is considered a model of the “recognition phase” of the homograft reaction, representing the initial recognition by, and subsequent proliferation of, lymphocytes responding to histocompatibility differences. In this “classical” MLC no attempt is made to identify individual factors responsible for histoincompatibility; rather, the total disparity between two individuals is measured. The way in which this measurement is made, the genetic control of reactivity, and the experimental evidence for the relationship of MLC reactivity to other forms of histocompatibility testing and to the actual fate of transplanted cells or organs, will be discussed in the sections which follow.
