Abstract
Late in 1968 a simple experiment was done. A commercial sample of salmon sperm DNA was denatured, annealed and examined in the EM. The results startled us: there were many rings to be found among the denatured chains and unidentifiable material. Bacterial and phage DNA’s did not produce these rings. Since that time we have produced these rings (called “slipped rings”) from every species of eukaryotic DNA that has come into our hands, including Drosophila polytene chromosome DNA. Prokaryotic DNA’s always failed to produce slipped rings. Such rings would be expected if chromosomal DNA contained a lot of tandem repetitions. The experiment was done to see if we could prove or disprove CALLAN’S suggestion that chromomeres were largely organized into tandemly-repeating sequences (Callan and Lloyd, 1960; Callan, 1967).
