Zusammenfassung
Ehe man die Struktur von Antikörpern untersuchen kann, muß man passende Methoden haben, um diese in hochgereinigter Form erhalten zu können. Für viele biologische und immunologische Zwecke ist die eigentliche Reinigung der Antikörper nicht so wichtig wie das Problem, sie so anzureichern, daß sie in größerer Konzentration als im Serum vorliegen. Dazu genügt es, konzentriertes γ-Immunglobulin durch Chromatographie an DEAE-Cellulose herzustellen, wobei man die Hauptfraktion nimmt, die bei Elution mit 0,005 oder 0,001 M Phosphatpuffer bei pH 8 nicht zurückgehalten wird. Oder man präcipitiert das Globulin durch Halbsättigung mit Ammonsulfat oder Natriumsulfat. Zur Trennung von γM- und γG-Immunglobulinen können weitere Fraktionierungen konzentrierter γ-Globulinlösungen mit Hilfe von Zonenelektrophorese, Gelfiltration an Sephadex oder Biogel, Dichtegradientenzentrifugation in Sucrose oder Kombinationen dieser Methoden durchgeführt werden. Diese Verfahren trennen gewöhnlich Antikörper von anderen Immunglobulinen derselben Klasse nicht ab.
