Zusammenfassung
Von wenigen neuen Techniken hat man je so wesentliche Verbesserungen bei der Diagnostik von Krankheiten erwartet wie von der Polymerasekettenreaktion (PCR). In der Tat handelt es sich bei der PCR um ein Nachweisverfahren von bisher nicht gekannter Empfindlichkeit. Mit der PCR läßt sich die sprichwörtliche Stecknadel im Heuhaufen finden. Der hohen Sensitivität liegt zugrunde, daß eine für das Nachzuweisende spezifische Nukleinsäuresequenz in meist bis zu 30 Zyklen, bestehend aus Denaturierung, Primeranlagerung und Doppelstrangsynthese erst millionenfach vermehrt wird — daß sozusagen mehr Stecknadeln als Heuhaufen vorhanden sind -, bevor dann der eigentliche Nachweis erfolgt.
