Zusammenfassung

Humaner rekombinanter von Willebrand-Faktor (rvWF) wird in CHO-Zellen mit rFurin koexprimiert, aus dem Zellkulturüberstand durch ein mehrstufiges chromatographisches Verfahren gereinigt und pharmazeutisch formuliert. Die biochemische, strukturelle und funktionelle Charakterisierung zeigt eine weitgehende Identität mit plasmatischem von Willebrand-Faktor, jedoch eine höhere Integrität [3]. Im Präparat sind Multimere hoher Molekülmasse vorhanden, vergleichbar mit jenen in humanem Normalplasma und Faktor VHI/von Willebrand-Faktorkomplexkonzentraten, die therapeutisch angewendet werden. In dieser Studie soll nun die weiterführende, funktionelle Charakterisierung des rekombinanten von Willebrand-Faktors in vitro und in vivo dargestellt werden.
