Zusammenfassung
Im colorectalen Carcinom können unterschiedliche Faktoren zu einer Akkumulation von cytosolischem β-Catenin führen: APC-Gen Mutationen [1] (in 80% aller sporadischen colorectalen Carcinome [2]), der Verlust von E-Cadherin [3], Mutationen im β-Catenin-Gen selbst [4] sowie die Überexpression der β-Catenin mRNA [5]. Akkumuliertes cytosolisches β-Catenin bildet mit Tcf/Lef-Proteinen einen Transkriptionsfaktor, der den letzten Schritt des Wnt/wingless Signaltransduktionsweges darstellt. Mehrer Hinweise sprechen dafür, daß die auf diesem Weg aktivierten Gene eine zentrale Rolle für die Coloncarcinogenese spielen könnten. Kürzlich wurde ein erstes Zielgen, c-myc, in HT 29 Zelle beschrieben [6]. Wir haben uns die Aufgabe gestellt, weitere Zielgene des β-Catenin-Tcf/Lef Komplexes zu identifizieren.
