Zusammenfassung
Laboruntersuchungen in der Medizinischen Genetik gehen auf die Arbeiten Sir Archibald Garrods zurück, der zu Beginn dieses Jahrhunderts seine Studien über seltene Stoffwechseldefekte wie z. B. die Alkaptonurie durchführte und in seiner Monographie Inborn Errors of Metabolism beschrieb. Garrod entwickelte das Konzept einer biochemischen Individualität, das besagt, daß individuelle Proteinvarianten für definierte Erkrankungen verantwortlich gemacht werden können. Der erste direkte Nachweis, daß Mutationen die Primärstruktur von Proteinen verändern, wurde 1949 von Pauling durch elektrophoretische Untersuchungen des Sichelzellhämoglobins erbracht. Zwei Jahre später konnte gezeigt werden, daß bei Patienten mit einer Sichelzellanämie im Hämoglobin ein Glutaminsäurerest gegen einen Valinrest ausgetauscht ist (Ingram (1956)).
