Abstract
Microsequencing of polypeptides by the Edman degradation technique can be performed either manually or by automated methods (Edman and Begg 1967; Edman and Henschen 1975). The degradation consists of three parts, coupling with 4-N, N′-dimethylaminoazobenzene-4′-isothiocyanate/phenylisothio-cyanate (DABITC/PITC), acid cleavage of the first amino acid as phenylthiazolinone from the peptide chain, and isomerization to the more stable phenylthiohydantoins. The stable derivatives are identified on thin-layer polyamide sheets (Edman and Henschen 1975) or by high performance liquid chromatography (HPLC) (Chang 1988).
