Zusammenfassung
Die Grundvoraussetzung dieser Methode ist das Vorhandensein einer primären Aminosäure am N-terminalen Ende des zu untersuchenden Polypeptides, d. h. die Methode versagt, wenn die N-terminale Aminosäure z. B. durch Methylierung oder Acetylierung in einem posttranslationalen Prozeß modifiziert worden ist. Wenn aber diese Grundvoraussetzung gegeben und die Aminosäurekette nicht verzweigt ist, steht der Anwendung der Methode nichts im Wege, sie ist dann das Mittel der Wahl, wenn der Nachweis erbracht werden soll, daß ein Polypeptid wirklich rein, d. h. nicht mit anderen Proteinen vergesellschaftet ist.
