Environmental Influence on Neurotrophic Gene Expression After Experimental Brain Infarction in the Rat

Summary

We have investigated whether post-ischemic housing in an activity-stimulating environment stimulates gene expression for brain-derived neurotrophic factor (BDNF), a neurotrophic factor known to stimulate brain plasticity in intact animals, in focal brain ischemia. From 30 h after a middle-cerebral-artery ligation, rats were housed in standard cages or in larger cages, allowing various activities. In situ hybridization was performed on brain from rats killed 2, 3, 7 and 12 days after the arterial ligation. Contrary to our hypothesis, rats in an enriched environment had significantly lower BDNF messenger RNA (mRNA) than rats in standard environment in the pert-infarct area, cortex contralateral to infarction and hippocampus. Determination of whether or not the lower BDNF mRNA levels in activity-stimulated rats corresponds to lower synthesis of BDNF is needed to evaluate the functional significance of the data.
