Zusammenfassung
Das atopische Ekzem stellt eine genetisch komplexe Erkrankung dar, bei der das Zusammenspiel mehrerer Gene und Genvarianten sowie der bedeutende Einfluß äußerer Faktoren die Aufklärung zugrundeliegender molekulargenetischer Defekte erschweren. Eine geeignete wissenschaftliche Vorgehensweise soll am Beispiel des IL4-Rezeptorgens aufgezeigt werden. Der Identifikation der chromosomalen Region 16p11.2/12 (als an die Krankheit gekoppeltem Genort) folgte die Identifikation des IL4-Rezeptorgens als Kandidatengen der Atopieentstehung. Mehrere häufige Polymorphismen im kodierenden Genabschnitt wurden isoliert und für einige ein Einfluß auf die Rezeptorfunktion nachgewiesen. Das Beispiel des IL4-Rezep-torgens soll auch verdeutlichen, daß nicht wenige Mutationen sondern vielmehr das Zusammenspiel einiger häufiger Genvarianten das individuelle Risiko definiert, an einem atopischen Ekzem zu erkranken.
