Abstract

Intracellular fluorescence measurements on flow cytometers can generate a variety of in situ data for physiological assays, functional assays, enzyme kinetic assays, as well as intracellular antigen detection. Many of these assays can be performed on live cells under normal physiological conditions. The stimulants or agonist can be then added in real time and the cell’s responses recorded to determine population dynamics. Geoffrey Osborne et al present an intracellular calcium flux measurement which has become a standard for many immunologists and cell biologists looking at cellular response to ligand binding or other stimuli. Mike Fox describes two different methods to measure intracellular pH. Norberg et al use the intracellular pH to measure apoptosis. Rybak and Murphy use a ligand acidification kinetic assay on several kinds of cells.
