Abstract

Myeloablative conditioning regimens are still required to achieve stable engraftment of allogeneic hematopoietic stem cells (HS). Here we show that preimplantation stage-derived rat embryonic stem cells (ES) can be injected intraportally into allogeneic hosts and survive without any supplementary conditioning. In addition, WKY-derived ES cells induce a state of tolerance for second-set transplanted heart allografts sharing the same donor haplotype (WKY). The underlying tolerance mechanism is in part based on constitutive expression of CD95 ligand, which allows the differentiation of ES cells into B cells and macrophages, hereby creating a partial mixed chimerism. Our results indicate a potential clinical application of ES cells as vectors of tolerance and as vehicles for gene transfer.
