Abstract

The influence of gadolinium chloride (GdCl3)-pre-treatment on the gene expression of pro-inflammatory (IL-1β, IL-6, TNF-α) and anti inflammatory genes (IL-1β receptor antagonist (IL-1RA), IL-10) in Kupffer cells derived from rats with an LPS induced septic shock was investigated using RT-PCR technique. Kupffer cells were harvested 1 h, 6 h, 12 h after LPS injection and a substantial reduction of IL-6, IL-10, and IL-1β, and a slightly decreased TNF-α as well as an approximately equal IL-1RA gene expression was observed at most time points compared to NaCl pre-treated control animals. These results indicate that GdCl3-blockade of Kupffer cells might not be suitable to treat a hyperinflammation since the expression of anti inflammatory acting genes was also depressed.
