MRI of experimental cerebral ischemia: response to NMDA antagonist therapy

Summary

MRI was used to assess the efficacy of the N-methyl-D-aspartate (NMDA) antagonists dextromethorphan and dextrorphan, for the treatment of cerebral ischemia in an animal model. Marked protective effects of treatment were seen by MRI and histologic measures (67% to 83% reduction in the lesion size, compared to controls). Relaxation time measurements showed that treatment reduced the prolongation of T1 and T2 in lesions. A close correlation was seen between high signal on T2-weighted images, histologic measures of ischemic neuronal damage, and reduced lesion blood flow. Meg-netic susceptibility effects of intravascular dysprosium-DTPA-BMA were seen on dynamic gradient recalled acquisition in the steady state (GRASS) images and T2-weighted spin-echo images. This contrast agent enhanced the early detection of ischemia on T2-weighted spin-echo images.
