Zusammenfassung
In den letzten Jahren ist es gelungen, mit Hilfe von Radioligand-Bindungsstudien adrenerge Rezeptoren direkt auf molekularer Ebene nachzuweisen (Übersicht bei Lefkowitz, 1978). Somit hat man zum ersten Mal die Möglichkeit, Rezeptor-Anzahl und -Empfindlichkeit direkt zu bestimmen. Dabei wurde es offensichtlich, daß die Anzahl (und damit Ansprechbarkeit) adrenerger Rezeptoren keine feste Größe ist, sondern durch eine Vielzahl von Pharmaka, Hormonen, physiologischen und pathologischen Zuständen reguliert werden kann. Solche Rezeptor-Veränderungen sind zunächst an einer Reihe von Tiermodellen studiert worden (Übersichten bei Hoffman und Lefkowitz, 1980; Barnes, 1981; Lefkowitz, 1982), während Studien am Menschen lange Zeit nicht möglich waren, da kein leicht zugängliches, adrenerge Rezeptoren tragendes, Gewebe bekannt war. Mit der Entdeckung, daß Thrombozyten α 2-Adrenozeptoren enthalten (Jakobs et al., 1976; Newman et al., 1978; Brodde et al., 1982d) hat sich jedoch die Möglichkeit ergeben, auch am Menschen α-Adrenozeptor-Veränderungen bei bestimmten Krankheitsbildern zu studieren.
