Zusammenfassung
In der Pathogenese der sklerotischen Gefäßwandprozesse spielt die Proliferation mesenchymaler Zellen aus Intima, Media und Adventitia eine Schlüsselrolle. Andererseits kommt es dabei durch lokale Endothelläsionen und Gerinnungsaktivierung zur Abscheidung von Mikrothromben, welche inkorporiert und organisiert werden. Es resultiert eine weitere Verdickung der Gefäßwand und somit ein Fortschreiten des arteriosklerotischen Prozesses (Duguid 1946). Gerinnungsaktivierung und Zellproliferation treten also bei der Arteriosklerose parallel zueinander auf. Unsere Untersuchungen galten dem Problem, ob und wie Gerinnung und gesteigerte Vermehrung von Gefäßwandzellen miteinander ursächlich verknüpft sind. Es sollte der Einfluß von Thrombin, Faktor XIII (F XIII), Kallikrein und Fibronectin auf das Proliferationsverhalten von Fibroblasten, Endothelzellen und glatten Muskelzellen analysiert werden. Weitere Untersuchungen galten dem Ziel, eine Vorstellung über die zugrundeliegenden Wirkmechanismen zu erhalten.
