Zusammenfassung
Wenn ein Organ oder Gewebe unter normalen Bedingungen eine biologisch hochwirksame Substanz enthält, so stellt sich die Frage nach ihrer physiologischen Bedeutung. Dabei ist entweder an einen Einfluß auf die Funktion des betreffenden Organs oder an eine Fernwirkung auf außerhalb davon gelegene Receptoren zu denken, und auch Kombinationen dieser beiden Möglichkeiten sind in Betracht zu ziehen. Diese Regel wurde bei der Bearbeitung von Renin zunächst nicht befolgt, und erst 50 Jahre nach seiner Entdeckung begann man, sich für seine Beteiligung an regulatorischen Vorgängen zu interessieren. Zwei Gründe sind für diese Verzögerung verantwortlich: erstens die blutdrucksteigernde Wirkung von Renin, die nicht nur seine hervorstechendste, sondern auch die am einfachsten nachweisbare Eigenschaft ist, und zweitens die Zusammenhänge zwischen Nierenerkrankungen und Hochdruck, die Veranlassung gaben, in einem pressorisch wirkenden Stoff renalen Ursprungs ein für die Pathogenese der Hypertension maßgebendes Agens zu sehen. Die Auffassung Volhards, daß bei bestimmten Formen von Hochdruck eine generelle Vasoconstriction auf humoralem Wege ausgelöst wird, wurde durch nahme des Reningehaltes in den Nieren einher, und die Reninaktivität im Plasma verläuft parallel dazu (Gross et al., 1965). Dabei ändert sich der Blutdruck im allgemeinen nicht. Bei der Ratte kann nach chronischer Salzzufuhr, besonders bei gleichzeitiger Gabe hoher Dosen von Cortexon oder Aldosteron, ein Hochdruck auftreten, wobei Renin aus den Nieren und aus dem Plasma verschwindet. Derartige Änderungen des Reningehaltes in den Nieren benötigen meist 2 bis 3 Wochen bis zu ihrer vollen Entwicklung, und die gleiche Zeit ist für ihre Rückbildung nach Absetzen der dafür verantwortlichen Behandlung erforderlich.
