Abstract
The antibiotic tirandamycin specifically inhibits transcription by interfering with the function of the bacterial enzyme RNA polymerase. The antibiotic is a 3-acyltetramic acid and thus structurally related to streptolydigin. Based on studies with synthetic analogs it is concluded that the tetramic acid nucleus is responsible for the recognition of RNA polymerase in conjunction with various specific substituent groups at positions 1, 3, and 5 of the tetramic acid.
