Abstract

In 1951 Kavanagh isolated from two basidiomycetes species, Pleurotus mutilus (Fr.) Sacc. and Pleurotus Passeckerianus Pilat, a substance which showed antibacterial activity. Kavanagh et al. (1951) named the crystalline material pleuromutilin. The antibacterial activity of this compound was modest and its spectrum largely confined to gram-positive organisms. Kavanagh and coworkers proposed for pleuromutilin the empirical formula C22H34O5; this was confirmed by subsequent chemical work. After a decade, the structural formula of this antibiotic was elucidated by both Arigoni’s group in Zürich (Naegeli, 1961; Arigoni, 1962) and by Birch and co-workers in Manchester (Birch etal., 1963; Birch etal., 1966). The sterochemical details of Formula I were established by Bonavia (1968) on the basis of X-ray and ORD data.
