Abstract
In recent years interest concerning morphogenetic processes have centred increasingly on the earliest stages of development. Mammalian ova have stayed at the margin of research of early development, although they represent the very object for investigation of the first processes of differentiation. The reason for this is no doubt the circumstance that procuring a sufficiently large number of mammalian ova at the period of fertilization and especially of cleavage encounters a whole series of technical obstacles which become multiplied when the specimens are prepared for electron microscopy.

The submicroscopic structure was studied on a total of 279 developmental stages of ova, comprising one-, two-, three-, four-, five-, and eight-cell ova, the morula and the blastocyst. The ova were obtained by flushing out the uterine tube and the uterine horn after the standard technique (Mazanec and Pliczka, 1960). Immediately after having been flushed out, the ova were fixed in a 1 % solution of osmiumtetroxide according to Zetterqvist (1956) at a pH of 7.6, in 1% OsO4 according to Dalton (1955) at pH 7.8, or in 1% glutaraldehyde in a phosphate or cacodylate buffer at pH 7.2 and postfixed with 1 % OsO4 in a phosphate or cacodylate buffer. To improve the osmotic properties 0.045 g/ml of sucrose was added to the fixative solution in some cases. After fixation, the specimens were quickly embedded in agar gel, the agar blocks were dehydrated and embedded in a mixture of methacrylates or Durcupan ACM. The sections made on the ultramicrotomes Reichert OMU 1 or Tesla BS 490, were stained with lead according to Karnovsky (1961) or with a 1% aqueous solution of uranyl acetate followed by lead citrate according to Reynolds (1963) (except the material fixed according to Dalton), and examined under the electron microscopes Tesla BS 242 D or Tesla BS 413 A.
