Zusammenfassung
1898 beschrieben Tigerstedt u. Bergmann [23] erstmals eine blutdrucksteigernde Substanz, die sie in wäßrigen Extrakten von normalen Kaninchennieren nachgewiesen hatten und Renin nannten. Die Autoren wiesen darauf hin, daß Renin bei der Entstehung der renalen Hypertonie von Bedeutung sein könnte. Mehr als 50 Jahre stand Renin nur im Mittelpunkt der Hochdruckforschung. Unter dem Einfluß von Volhard, der bei seiner klinischen Einteilung in roten und blassen Hochdruck als Ursache für letzteren ein humorales Agens renalen Ursprungs (Renin ?) annahm, gelang es 1930 Hartwich [11] bei Hunden durch Drosselung der Blutzufuhr zu einer Niere eine vorübergehende Blutdrucksteigerung zu erzielen. Goldblatt [6] konnte 1934 in systematischen Untersuchungen zeigen, daß durch Herabsetzung der Durchblutung einer Niere eine Hypertonie erzeugt werden kann. Weitere Untersuchungen führten zum Nachweis, daß Renin nicht selbst blutdrucksteigernd, sondern als Enzym wirkt, das aus dem Substrat Angioten-sinogen das vasopressive Angiotensin freisetzt [1, 14]. Nach der Identifizierung [4, 18] von Angiotensin (1956) gelang 1957 seine Synthese [3, 16]. Erst Gross (1958) [7] zeigte durch seine Untersuchungen an Ratten, daß dem in normalen Nieren vorkommenden Renin eine physiologische Bedeutung zukommt. Inzwischen sind mehrere physiologische Funktionen für das Renin-Angiotensinsystem bekannt, ein direkter Zusammenhang zwischen diesem System und der renalen Hypertonie jedoch immer noch nicht bewiesen.
