Daunomycin and Related Antibiotics

Abstract

Daunomycin (Da) is a metabolite of Streptomyces peucetius (Grein et al., 1963) which strongly inhibits a variety of experimental tumors (Di Marco et al., 1964b). The antibiotic substance is a glycoside (Hydrochloride: C27H29NO10 · HCl) which by acid hydrolysis yields an aglycone, daunomycinone (Arcamone et al., 4964b) and a new aminosugar, daunosamine (Arcamone et al., 4964a). Dubost et al. (4963) have also described an antibiotic, rubidomycin, whose physicochemical characteristics correspond to those of daunomycin. Daunomycin is closely related to the anthracyclines. Following Brockmann’s suggestion (4963), those antibiotics that contained a tetrahydrotetracenquinone chromophore linked to a sugar should all be included in the anthracycline group. The chromophores of such antibiotics have different substituents on the nucleus (Fig. 1 and Table 1).
