Zusammenfassung
Der Alkaptonurie (OMIM #203500) liegt eine autosomal-rezessiv vererbte Aktivitätsminderung des Enzyms Homogentisinsäure-1,2-Dioxygenase (HGD) zugrunde. Dadurch kann Homogentisinsäure, ein Zwischenprodukt in den Abbauwegen der aromatischen Aminosäuren Phenylalanin und Tyrosin, nicht weiter abgebaut werden und akkumuliert im Organismus. Die Krankheitssymptome werden durch die pathologische Anhäufung der Homogentisinsäure (HGA) verursacht. Hohe Konzentrationen von Homogentisinsäure im Urin führen zu dessen Dunkelfärbung. Die chemische Grundlage der Dunkelverfärbung wurde bereits im 19. Jahrhundert aufgeklärt. Die Alkaptonurie wurde 1902 von Garrod als klinische Entität und vermutliche erbliche Stoffwechselstörung beschrieben
