Development of IgY-Based Sandwich Enzyme-Linked Immunosorbent Assay for the Detection of Vibrio parahaemolyticus

Abstract

Vibrio parahaemolyticus is one of the most important food-borne pathogen which can cause serious infection in human. One kind of sandwich ELISA method was developed in this paper to detect V. parahaemolyticus rapidly and economically, using anti- V. parahaemolyticus IgY as capture antibody and rabbit anti- V. parahaemolyticus IgG as detection antibody. The specific IgY was harvested from egg yolk of immunized hens, then was purified by ammonium sulfate precipitation, Sephadex G-25 gel chromatography and DEAE-Sehparose FF ion exchange chromatography. The established assay had a limit of detection of 2.5×104 cfu/ml. The cross reactivity testing showed a high specificity for V. parahaemolyticus detection and other observed bacteria were found to be non-reactive. The IgY coated microtiter plate had high stability. Results suggest that IgY-based sandwich ELISA in this study provides a simple and convenient preparation for the development of immunoassay-kit and rapid detection.
