Abstract

Transgenic animals have become an important tool that has helped to shape our current understanding of the mechanisms of lipoprotein metabolism. They have also proved invaluable for the evaluation of therapeutic strategies directed against disorders of lipid homeostasis and related diseases such as atherosclerosis, obesity, and Alzheimer’s disease. In this chapter, we will briefly outline the biochemical and physiological basis of cellular and systemic lipoprotein metabolism and review selected examples how genetically modified animals have aided us in dissecting the molecular function of the genes that regulate lipoprotein transport in the context of the whole organism.
