Zusammenfassung
Die Kenntnis und die Beachtung pharmakokinetischer Prinzipien stellen eine der Grundlagen der Arzneimitteltherapie dar. Zusätzlich können durch die Bestimmung der Konzentrationen der Arzneimittel und der gebildeten Abbauprodukte die Voraussetzungen für die Erstellung und die differenzierte Betrachtung von Konzentrations-Wirkungs-Beziehungen von Arzneimitteln unter Einbeziehung ihrer Metabolite geschaffen werden. Die Integration von Pharmakokinetik und Pharmakodynamik ergänzt durch die Erkenntnisse über den Einfluss genetischer Aspekte im Arzneimittelstoffwechsel oder von spezifischen Transportprozessen hat mit dazu beigetragen, dass Arzneimittel mit geringer therapeutischer Breite mit größerer therapeutischer Sicherheit auch bei Patienten aus Risikogruppen (multi- morbide Patienten, Patienten mit Erkrankungen der Eliminationsorgane) angewandt werden können.
