Modern Strategies in the Design of Antimicrobial Agents

Abstract

Several statistical and graphical methods and systems have been developed during recent decades to assist in a more rational approach to drug design. Together with an increased knowledge of molecular biology and the development of simple, suitable in vitro test systems, a better understanding of the various steps in drug action and their structural dependence has been achieved. The newly developed computerized techniques are especially helpful for the systematic development of new antibacterial agents. Three major applications of these techniques are reviewed: (1) Lead generation by computer graphics, molecular modelling and three-dimensional design; (2) Rationalization in the optimization procedure of lead compounds by quantitative structure-activity relationship (QSAR) techniques; and (3) Comparison of various biological activity data derived from test systems by multivariate data analysis to avoid unnecessary testing and to separate pharmacodynamic and pharmacokinetic effects.
