Basic Confocal Microscopy

Abstract

In the opening paragraph of this text, we noted that often students and technologists use a confocal microscope to collect images without a full understanding of how the system works. Unfortunately, an additional factor affecting image collection is that often students and technologists are given a preconceived idea of what the final image or data should look like. With the variety of mechanisms available on a confocal system for changing the appearance of an image observed through a microscope, it is essential that the user does not manipulate settings to match expected outcomes concerning their data. This chapter provides 12 guiding principles that need to be addressed when collecting and processing digital images with confocal and other microscopes and a list of resources available for assistance with confocal imaging.
