The Inflammatory Networks and Dynamical Patterns in Psychoneuroimmunology

Abstract

Systemic PNI profiles can be constructed with the identification of inflammatory biomarkers, cytokine networks, and immune-brain-behavior communications. Such profiles can be useful for the discovery of potential therapeutic targets in personalized, systems, and dynamical medicine. The elucidation of the common systemic biomarkers such as the inflammatory pathways in different diseases may help make the progress from the disease-centered medicine to human-centered medicine. At the molecular and cellular levels, the networks of cytokines and other molecules including neurotransmitters and neuropeptides mediate the interconnections among the nervous, endocrine, and immune systems. At the system level, the complex communications such as those between the central nervous system (CNS) and the peripheral immune system are essential in emotionality and the regulations of mood, motivation, and alarm. The cytokine pathways may convey the communications between neurons and glial cells, with the impacts on the hypothalamic–pituitary–adrenal (HPA) axis. Such impacts may result in neuropsychiatric dysfunctions showing as anxiety, depression, anorexia, fatigue, cognitive problems, and sleep disorders. The dynamical studies of the inflammatory biomarkers and relevant pathways need to become a high priority in systemic PNI profiling with their key roles in the wide range of problems including mood disorders and schizophrenia. Because of the importance of the networks such as the BDNF-TrkB signaling pathways, they have been suggested as the potential treatment targets for inflammation-associated disorders such as depression.
