Abstract
This chapter describes a method for preparing plant cells and tissues for whole-mount immunofluorescence microscopy. The protocol consists basically of four steps: fixation, partial enzymatic removal of cell walls, permeabilization of plasma membrane, and immunofluorescent staining. This protocol significantly shortens the time needed for specimen preparation as it does not require embedding and sectioning of cells and tissues. When stained samples are studied using a confocal laser scanning microscope (CLSM), three-dimensional distribution of the targeted organelle and antigen can be discerned. This technique augments the conventional immuno- and fluorescent staining procedures.
