Abstract

The insulin receptor is an integral plasma membrane glycoprotein of most cells. It consists of 2 subunits linked by disulfide bonds; the alpha and beta subunits of the receptor are synthesized by way of a single chain proreceptor which is cleaved and further processed, by the addition of complex carbohydrates, prior to insertion into the plasma membrane49. Recently a cDNA encoding the proreceptor has been cloned and the protein sequence of the receptor elucidated29,85. The alpha subunit of the receptor is the major binding component and the beta subunit is a tyrosine specific protein kinase which itself can be autophosphorylated.
