Abstract

Although the endothelium is considered the main site of production of the endothelins, many other cell types in the lung are capable of endothelin synthesis, implying numerous potential roles for endothelins in lung biology. The endothelins were characterised originally as powerful vasoconstrictors, however, it is becoming clear that their effects may vary depending on the predominant receptor subtype present and the initial tone of the vascular bed. In addition, endothelins may be mitogenic and chemotactic for lung fibroblasts and smooth muscle cells derived from the airways or pulmonary circulation. Furthermore, their presence during development raises the possibility that these peptides may contribute to growth of the normal lung in addition to pathological remodelling of the airways and lung vasculature.
