Summary

In gallamine-immobilized and artificially ventilated rats, the dose-reponse curves of M-agonists and AntiChE rather than GABA or glycine receptor antagonists inducing EEG seizures shifted leftward after the brain nAChR was desensitized by acute or chronic nicotine pretreatment. This phenomenon could be prevented by N-antagonist mecamylamine.Similar results were obtained in acute nicotine tolerant mice and rabbits.In addition, the effect of M-agonist arecoline for producing brain mAChR down-regulation was potentiated in acute nicotine tolerant rats. Apparent dissociation constant values of brain mAChR high-affinity site to its agonists arecoline and oxotremorine were decreased in the presence of nicotine. It is concluded that brain nAChR desensitization as a result of repeated nicotine pretreatment selectively increases the sensitivity of brain mAChR high affinity binding sites to its agonists.
