Summary
In clonal PC12 cells (differentiated with nerve growth factor), nicotine (100 µmol/1) induces exocytotic release of 3H-noradrenaline, since release (1) depends absolutely on the presence of extracellular calcium and (2) is associated with release of neuropeptide Y. The tricyclic antidepressant desipramine potently inhibits nicotine-induced noradrenaline release. However, desipramine is a much less potent inhibitor when release is elicited by the calcium ionophore A23187 or by potassium-induced depolarization. Desipramine most probably blocks the nicotinic acetylcholine receptor or the receptor-related cation channel.
