Summary
Nicotine (10 μM) induced a marked elevation of the immediate early gene mRNAs c-fos, c-jun, and jun-B. Maximally elevated levels for c-fos mRNA (about 100-fold) were obtained after 20 min. c-jun and jun-B mRNAs were increased 3- to 4-fold 60 min after nicotine addition. In contrast, the mRNA levels coding for proenkephalin showed, after an apparent lag-phase, a delayed (up to 4-fold) induction between 4 and 24 hours. The expression of the proenkephalin reporter gene PENKCAT-153/+53, containing 153 nucleotides of upstream promotor sequences, was increased about twofold by nicotine or by cotransfection of an expression vector for c-fos, which also enhances expression of PENKCAT(CRE2)2, indicating that nicotine may induce proenkephalin via c-Fos which binds at the ENKCRE-2 element.
