Abstract
Myocardial infarction, stroke, and pulmonary embolism are among the leading causes of mortality and morbidity in western countries [1–4]. These disorders have, as their underlying etiology, the occlusion of blood vessels by a clot (thrombosis). One approach to the treatment of thrombosis is the pharmacological dissolution of the clot (thrombolysis), by substances that activate the fibrinolytic system. This system leads to the conversion of the inactive proenzyme plasminogen, to plasmin. Plasmin degrades fibrin into soluble degradation products. To clarify the steps of this complex lytic mechanism, an introduction to the biochemistry and physiology of fibrinolysis and of its components will be provided. This is important to a better understanding of the mechanisms of the effect of currently available thrombolytic agents as well as to newer achievements in the area of thrombolysis.
