Abstract
The extreme sensitivity of diphtheria toxin production to the iron concentration of the medium was clearly shown by Pappenheimer and Johnson [1]. Iron level of the medium, therefore, has to be carefully controlled whether diphtheria toxin is produced using a surface or a submerged culture. However, the mechanism underlying the inhibition of toxin production by iron is not yet known. We have employed a purely synthetic medium, developed by us, to study the effect of iron on the bacillus as well as on toxin production. On the basis of the observations made, we have also put forward a hypothesis to explain the mechanism.
