Abstract

The biodisposition and metabolism of ibuprofen are important for understanding its applications, mode of actions and safety in various inflammatory and painful conditions where it is widely used. Ibuprofen is metabolised via oxidative—cytochrome P450 2C9, 2C19 and 2C8 (phase I)—and conjugation (phase II) pathways. The drug has a relatively short plasma elimination half-life and this is a feature in relation to its relative safety. The drug rapidly accumulates in synovial fluids and other compartments where it acts to control pain and inflammation. The pharmacokinetics of ibuprofen is closely related to its therapeutic effects in adults and children. There is relatively little variation in pharmacokinetics of ibuprofen in different inflammatory conditions, with relatively limited increases in plasma concentrations in moderate liver and renal diseases.
