Résumé
Présent chez la plupart des mammifères, le gène codant pour le BNP est situé sur le chromosome 1 (1p36.2) et se compose de 3 exons et de 2 introns chez l’Homme. Ce gène code une protéine de 134 acides aminés (AA), le pré-proBNP1–134. Ce précurseur est transformé par clivage enzymatique de son peptide signal (26 AA) en proBNP1–108 (108 AA), une glycoprotéine O-glycosylée. Une nouvelle phase de maturation nécessitant l’action de protéases, la furine et la corine, génère de façon équimoléculaire d’une part, une partie C terminale, une holoprotvine, le BNP1–32 (32AA) et, d’autre part, une partie N terminale, glycoprotéine de 76 AA, le NT-proBNP1–76, sans action physiologique connue à ce jour.
