Classification of T-cell and NK-cell Malignancies

Abstract/Scope of Chapter

This chapter provides an overview of mature T-cell malignancies, including those presenting in lymph nodes (e.g., angioimmunoblastic T-cell lymphoma), extranodal sites (e.g., hepatosplenic T-cell lymphoma), and leukemias (e.g., Sézary syndrome). The role of the two prototypic mature T-cell oncogenes, ALK kinase in anaplastic large cell lymphoma and TCL1 in T-cell prolymphocytic leukemia, is discussed. A general model for T-cell tumor initiation involving canonical and aberrant signaling through the T-cell receptor is presented. The clinical presentation and morphologic range of NK-cell malignancies are also discussed.
