IGF Binding Protein Proteolysis in Various Clinical States

Abstract

The insulin-like growth factors (IGFs) are generally present throughout the body at concentrations far higher than are required for maximal stimulation of their cell receptors. These high IGF concentrations are maintained by the six IGF binding proteins (IGFBPs), which are ubiquitously present in various combinations, generally at concentrations in excess of the IGFs. The affinities with which the IGFBPs bind the IGFs are much higher than those for the type 1 IGF receptor, which mediates most of their cellular actions. Factors that could lower the affinities of the IGFBPs may therefore play a critical role in regulating IGF availability. A general mechanism whereby this can be achieved is the limited proteolysis of the IGFBPs. If each IGFBP has its own specific, tightly regulated protease, then this could represent a very sophisticated mechanism to control IGF bioavailability.
