Abstract

The molecular era for the insulin-like growth factors (IGFs) began in 1978 with the purification and amino acid sequencing of human IGF-I and IGF-II (1,2). During the past 20 yr, IGF proteins have been characterized from species as diverse as humans, chickens, frogs, and sharks, with the bulk of the information being obtained during the last decade through cDNA cloning. Although in most species IGF-I and IGF-II are small single-chain peptides of 70 and 67 amino acids, respectively, molecular cloning has revealed a remarkable complexity and diversity in both IGFs and in their precursor proteins, mRNAs, and genes. This diversity is all the more surprising because the genes and mRNAs encoding insulin, the other major member of the insulin—IGF gene family, are relatively simple (3). The focus of this chapter is to summarize basic information on structural aspects of IGF-I and IGF-II and their genes. Readers are referred to other chapters for discussions of IGF gene regulation, and to previous reviews of IGF protein and gene structure and function (4–6).
