What Have We Learned from Guideline Studies?

Abstract

The goal of this chapter is to summarize the available developmental neurotoxicology data and to ascertain the value of developmental neurotoxicity as currently evaluated by the USEPA in the identification of human risk. A whole host of scientific methodologies have been used to evaluate nervous-system damage and it is beyond the scope of this chapter to provide a comprehensive overview of the entire body of research in the field of developmental neurotoxicology. Instead, the findings presented here will focus on developmental neurotoxicology studies executed in general accordance with the most recent US Environmental Protection Agency (EPA) Office of Prevention, Pesticides and Toxic Substances (OPPTS) Guideline 870.6300 (August 1998) (1). While our discussion revolves around studies executed in agreement with the OPPTS testing paradigm, the compounds reviewed are not limited to those under EPA regulatory jurisdiction. The considerable scientific literature available regarding investigation of the potential developmental neurotoxicity of pharmaceuticals and drugs of abuse (2) will also be discussed. However, by focusing on studies performed in general agreement with regulatory guidelines, we intend to assess the confidence in our ability to identify potential hazards to the developing nervous system.
