Summary

Aspects of the use of overpressured layer chromatography (OPLC) as an instrumentalised version of planar liquid chromatography, suitable for both analytical and preparative off-line and on-line separations, are described.

For example the eluent strength and solvent composition considerably influence the applicability of off-line OPLC owing to the demixing of the solvent during chromatography. The disturbing effect due to this demixing phenomenon can be reduced or eliminated if the differences in solvent strength between the eluent constitutents are kept to a minimum.

There is a strong correlation between dry and wet OPLC separations regarding retention characteristics, which offers a new prospect for the modelling of high performance liquid chromatography.

In the case of preparative on-line separation and detection prewetting the stationary phase from outlet side can eliminate disturbances which otherwise adversly affect detection.
