Potent Mitochondrial Inhibitors with Diverse Applications

Abstract

Annonaceous acetogenins are a relatively new, but rapidly growing, class of compounds. Since the first of them, uvaricin, was reported in 1982,1 more than 160 compounds have been published. These natural products have attracted considerable interest due to their diversity of bioactivities. Most acetogenins are potently bioactive and offer exciting potential as new antitumor, immunosuppressive, pesticidal, antiprotozoal, antifeedant, anthelmintic, and antimicrobial agents.2-7 Chemically, the Annonaceous acetogenins seem to be derivatives of C32 or C34 fatty acids which are combined with a 2-propanol unit at C-2; the propanol is incorporated into a 2,4-disubstituted γ-lactone which can assume several different forms. Usually, one, two, or three tetrahydrofuran (Thf) rings exist in the middle of the long hydrocarbon chain, which often contains a number of oxygenated moieties (hydroxyls, acetoxyls, ketones, epoxides), and/or a double bond.
