Abstract
Two-dimensional gels can provide quantitative information about the levels of expression and degree of modification for thousands of proteins from biological samples. However, obtaining that information is a difficult task involving 1) preparation of reproducible two-dimensional gels, 2) detection and quantitation of protein spots from complex gel images, 3) comparison of multiple gel images through matching techniques, 4) identification of the protein spots, and 5) characterization of protein modifications. At the QUEST Protein Database Center we have developed and refined methods for all aspects of 2D gel production and analysis, and we have used the QUEST system to build protein databases for rat and mouse cells. These databases have been explored quantitatively using cluster analysis to find proteins with similar expression patterns. These databases, and a potentially powerful new method for identification of protein spots in two-dimensional gels, are presented below.
