Abstract

During the past few years, developments in mass spectrometry have taken place that have had considerable impact on protein sequencing methodology. Fast atom (or ion) bombardment (FAB) ionization (Barber et al., 1981) has made it possible to determine the molecular weights of peptides and small proteins (Mr < 15 kDa) with an accuracy of ± 0.01% (i.e., ±0.1 to ± 1.5 Da), depending on molecular weight. More recently, the introduction of matrix-assisted laser desorption (MALD) (Karas and Hillenkamp, 1988) and electrospray (ES) (Meng et al., 1988) ionization have extended the range to hundreds of kilodaltons with a similar accuracy (0.01–0.1%).
