Age Influences the Synergy between Butyrylcholinesterase K Variant and Apolipoprotein E ε4 in Late-Onset Alzheimer’s Disease

Abstract

The presence of the K allele of butyrylcholinesterase (BCHE-K) gave an odds ratio of confirmed late-onset (>65y) Alzheimer’s disease of 2.15 (95% CI: 1.1–4.25) (74 cases, 104 controls). The association was confined to carriers of apolipoprotein E ε4 (APOEε4), in whom the BCHE-K allelic odds ratio of confirmed late-onset AD was 6.9 (1.65–29). When the cohort was stratified by age, significant results were only found in the >75y group, in whom the synergy factor of AD (see box) between BCHE-K and APOEε4 was 26.8 (p=0.02). The BCHE-K allele frequency in APOEε4-positive AD cases increased from 0.07 in those <65y to 0.13 in the 65–75y group and 0.24 in those >75y (p=0.01). These findings suggest strong synergy between BCHE-K and APOEε4 in the risk of AD in subjects >75y.
