Summary
The use of proton (1H) NMR as a method for studying the solid phase extraction (SPE) of biofluids is described with particular reference to studies undertaken on human urine. The extraction of both control urine and samples containing ibuprofen or paracetamol metabolites was used to investigate the extraction properties of a variety of phases for both endogenous compounds and drug metabolites. The phases studied included C18, β-cyclodextrin, aminopropyl, strong anion and strong cation exchange materials (SAX and SCX) and a multimodal phase exhibiting reversed-phase, SAX and SCX properties. In addition to enabling the study of the extraction properties of the sample components NMR also revealed the presence of phase-related material in the eluates from some SPE phrases.
The advantages of 1H NMR as a multiparametric “universal” detector for this type of investigation are contrasted with other general means of detection such as UV spectroscopy.
